Estudio preliminar del mecanismo de apagamiento de la fluorescencia de la lisozima por tetrakis-para-carboxifenil porfirina

Las interacciones moleculares son procesos centrales que regulan la vasta mayoría de reacciones catabólicas y metabólicas en los organismos vivos. Así mismo, el estudio de las interacciones intermoleculares proteína-ligando forman parte sustancial en las primeras etapas del desarrollo de nuevos agentes farmacéuticos. En esta investigación se sintetizó la tetrakis-para-carboxifenil porfirina, H₂T(p-COOH)PP, y se estudió su interacción con lisozima por métodos espectroscópicos: UV- visible y fluorescencia. Se encontró que la especie H₂T(p-COOH)PP no tiende a formar agregados, ni a protonarse en medio acuoso. Se observó el apagamiento intrínseco de la fluorescencia de la lisozima en presencia de H₂T(p-COOH)PP, el análisis a diferentes temperaturas usando el método de Stern-Volmer, reveló que el apagamiento es principalmente de carácter dinámico, por lo que se estableció que no existe un contacto importante para que se genere una interacción no covalente estable entre lisozima y H₂T(p-COOH)PP. Molecular interactions are central processes and mediate the majority of catabolic and metabolic reactions in living organisms. Therefore, the study of intermolecular interactions protein-ligand is an essential part for the early stages of the development of new pharmaceutical compounds. In this work, tetrakis-para-carboxyphenil porphyrin was synthetized and its interaction with lysozyme by spectroscopic methods, UV-vis and fluorescence, was studied. Aggregation or protonation of H₂T(p-COOH)PP did not tend to occur in aqueous medium. Intrinsic adhesion of fluorescence of lysozyme was observed in the presence of H₂T(p¬COOH) and the analysis at different temperatures using the Stern-Volmer method, revealed that H₂T[p-COOH)PP causes fluorescence quenching of lysozyme through a dynamic quenching process. Consequently, it was determined that it does not form a stable non-covalent interaction with lysozyme.
