Glucose-6-phosphate dehydrogenase (G6PD) is a vital enzyme that plays a crucial role in the production of NADPH. It is essential for the survival of red blood cells (RBC) by protecting them from oxidative damage and preventing hemolysis. Therefore, G6PD deficiency is a disease that affects RBC function. This deficiency induces neonatal jaundice and hemolytic anemia. Up to 230 G6PD variants have been identified; however, not all have been characterized. In this work, we report the characterization of three pathogenic G6PD variants (Tomah, Kangman, and Madrid), class A, with catalytic activity in erythrocytes less than 10%, which induce severe clinical manifestations in patients. Therefore, it is important to characterize these variants and understand their phenotype. The mutation is located at the same codon and alters Gly385. The variants were cloned, expressed, purified, and biochemically characterized, and complemented with in silico studies. All three variants were found to have lower affinity for G6P and NADP+, and lower thermal stability. Molecular dynamics simulations showed that the variants exhibit alterations particularly in the structural NADP+ binding site.
